During the development of the immune system, lymphoid tissue-inducing cells (LTi4) with CD4 markers are critical for the formation of lymph nodes and other secondary lymphoid organs. These cells are different from another congenital lymphocyte type ILC4neg that lacks CD4. The researchers found that retinoic acid affects the differentiation of these induced cells. They fed a diet rich in retinoic acid to pregnant mice and found that the ratio of LTi4 cells to ILC4neg cells in the embryos increased. Inhibition of retinoic acid signals has the opposite effect. The retinoic acid receptor-mutated embryos showed a decrease in the percentage of cells induced compared to the control embryos. The researchers thus suggested that vitamin A is essential for the differentiation of these LTi4 cells.
